The Roles of the Malic Enzymes of Rhizobium (Sinorhizobium) Meliloti in Symbiotic Nitrogen Fixation

dc.description.abstractThe genome of ๐˜™. ๐˜ฎ๐˜ฆ๐˜ญ๐˜ช๐˜ญ๐˜ฐ๐˜ต๐˜ช contains two genes for malic enzymes. One uses NADโบ as a cofactor (๐˜ฅ๐˜ฎ๐˜ฆ) and one utilizes NADPโบ (๐˜ต๐˜ฎ๐˜ฆ). The two enzymes have been purified and the genes cloned and sequenced. Loss of TME enzyme function gives no detectable phenotype in either ๐˜™. ๐˜ฎ๐˜ฆ๐˜ญ๐˜ช๐˜ญ๐˜ฐ๐˜ต๐˜ช grown in culture or in bacteroids. Loss of DME function gives no detectable phenotype in ๐˜™. ๐˜ฎ๐˜ฆ๐˜ญ๐˜ช๐˜ญ๐˜ฐ๐˜ต๐˜ช grown in culture but does result in bacteroids that are unable to fix nitrogen (Fixโป). Expression of ๐˜ต๐˜ฎ๐˜ฆ is reduced in bacteroids whereas ๐˜ฅ๐˜ฎ๐˜ฆ expression remains unchanged. In order to overexpress ๐˜ต๐˜ฎ๐˜ฆ in bacteroids a fusion gene was constructed with the ๐˜ฅ๐˜ฎ๐˜ฆ promoter driving expression of the ๐˜ต๐˜ฎ๐˜ฆ structural gene (๐˜ฅ๐˜ต๐˜ฎ๐˜ฆ). The ๐˜ฅ๐˜ต๐˜ฎ๐˜ฆ gene was expressed and functional in ๐˜™. ๐˜ฎ๐˜ฆ๐˜ญ๐˜ช๐˜ญ๐˜ฐ๐˜ต๐˜ช cells grown in culture, but alfalfa plants inoculated with strains expressing only the ๐˜ฅ๐˜ต๐˜ฎ๐˜ฆ gene were Fixโป. In addition the NADโบ-dependent malic enzyme gene from ๐˜š๐˜ต๐˜ณ๐˜ฆ๐˜ฑ๐˜ต๐˜ฐ๐˜ค๐˜ฐ๐˜ค๐˜ค๐˜ถ๐˜ด ๐˜ฃ๐˜ฐ๐˜ท๐˜ช๐˜ด (๐˜ฎ๐˜ข๐˜ฆ๐˜Œ) was similarly cloned downstream of the ๐˜ฅ๐˜ฎ๐˜ฆ promoter. The fusion gene ๐˜ฅ๐˜ฎ๐˜ข๐˜ฆ๐˜Œ was expressed in ๐˜™. ๐˜ฎ๐˜ฆ๐˜ญ๐˜ช๐˜ญ๐˜ฐ๐˜ต๐˜ช cells grown in culture, surprisingly plants inoculated with strains expressing only the ๐˜ฅ๐˜ฎ๐˜ข๐˜ฆ๐˜Œ gene showed a Fixโป phenotype. A truncated ๐˜ฅ๐˜ฎ๐˜ฆ gene was constructed which contained only the N-terminal, malic enzyme domain of the protein (๐˜ฅ๐˜ฎ๐˜ฆฮ”๐˜—๐˜ด๐˜ต). The truncated enzyme was expressed and active in ๐˜™. ๐˜ฎ๐˜ฆ๐˜ญ๐˜ช๐˜ญ๐˜ฐ๐˜ต๐˜ช cells grown in culture and gave a Fixโบ phenotype when inoculated onto alfalfa plants.en_US
