Abstract
The genome of ๐. ๐ฎ๐ฆ๐ญ๐ช๐ญ๐ฐ๐ต๐ช contains two genes for malic enzymes. One uses NADโบ as a cofactor (๐ฅ๐ฎ๐ฆ) and one utilizes NADPโบ (๐ต๐ฎ๐ฆ). The two enzymes have been purified and the genes cloned and sequenced. Loss of TME enzyme function gives no detectable phenotype in either ๐. ๐ฎ๐ฆ๐ญ๐ช๐ญ๐ฐ๐ต๐ช grown in culture or in bacteroids. Loss of DME function gives no detectable phenotype in ๐. ๐ฎ๐ฆ๐ญ๐ช๐ญ๐ฐ๐ต๐ช grown in culture but does result in bacteroids that are unable to fix nitrogen (Fixโป). Expression of ๐ต๐ฎ๐ฆ is reduced in bacteroids whereas ๐ฅ๐ฎ๐ฆ expression remains unchanged. In order to overexpress ๐ต๐ฎ๐ฆ in bacteroids a fusion gene was constructed with the ๐ฅ๐ฎ๐ฆ promoter driving expression of the ๐ต๐ฎ๐ฆ structural gene (๐ฅ๐ต๐ฎ๐ฆ). The ๐ฅ๐ต๐ฎ๐ฆ gene was expressed and functional in ๐. ๐ฎ๐ฆ๐ญ๐ช๐ญ๐ฐ๐ต๐ช cells grown in culture, but alfalfa plants inoculated with strains expressing only the ๐ฅ๐ต๐ฎ๐ฆ gene were Fixโป. In addition the NADโบ-dependent malic enzyme gene from ๐๐ต๐ณ๐ฆ๐ฑ๐ต๐ฐ๐ค๐ฐ๐ค๐ค๐ถ๐ด ๐ฃ๐ฐ๐ท๐ช๐ด (๐ฎ๐ข๐ฆ๐) was similarly cloned downstream of the ๐ฅ๐ฎ๐ฆ promoter. The fusion gene ๐ฅ๐ฎ๐ข๐ฆ๐ was expressed in ๐. ๐ฎ๐ฆ๐ญ๐ช๐ญ๐ฐ๐ต๐ช cells grown in culture, surprisingly plants inoculated with strains expressing only the ๐ฅ๐ฎ๐ข๐ฆ๐ gene showed a Fixโป phenotype. A truncated ๐ฅ๐ฎ๐ฆ gene was constructed which contained only the N-terminal, malic enzyme domain of the protein (๐ฅ๐ฎ๐ฆฮ๐๐ด๐ต). The truncated enzyme was expressed and active in ๐. ๐ฎ๐ฆ๐ญ๐ช๐ญ๐ฐ๐ต๐ช cells grown in culture and gave a Fixโบ phenotype when inoculated onto alfalfa plants.
